A user's effective access is the full set of things they can do after Salesforce combines their one profile with every permission set and permission set group assigned to them. Profiles and permission sets add object and field permissions; they do not take permissions away. A muting permission set can turn selected permissions off inside one permission set group, but the user still has a permission if their profile, a standalone permission set, or another group also grants it.

Core information

A user gets exactly one profile, usually aligned to a job function such as sales representative or administrator, and can also receive multiple permission sets stacked on top of it. Permission sets extend functional access without changing that profile, and you can assign them to users regardless of which profile they already have.

More details and nuances

Profiles, permission sets, and permission set groups can grant access but cannot deny it. To revoke a permission you must remove every instance of it from the user, by assigning a different profile, removing permission set and group assignments, cloning those assignments and turning the permission off on the clone, or muting it inside a group. Disabling the permission on a shared profile or set also removes it from every other user assigned that same profile or set.
