Sharing to a role and sharing to a role plus subordinates are two different targets for the same extra access. A role is a named level in Salesforce's data-access hierarchy; Roles gives the grant only to people assigned to that level, while a subordinates option also includes everyone in roles beneath it. Use the narrow target when only that job level should see the records or folder, and the broader target when the whole branch under that role should.

Roles as a sharing or group target includes every user assigned to that exact role and excludes site and portal roles.

Access from a sharing rule also rolls up to superiors for standard objects, and for custom objects when Grant Access Using Hierarchies is selected; turning that setting off for a custom object stops the roll-up, so only the owner and users you grant access to can see those records.
